Cell lines and clearing approaches: a single-cell level 3D light-sheet fluorescence microscopy dataset of multicellular spheroids

Nowadays, three dimensional (3D) cell cultures are widely used in the biological laboratories and several optical clearing approaches have been proposed to visualize individual cells deepest layers of cancer multicellular spheroids. However, defining most appropriate approach for different lines is an open issue due lack a gold standard quantitative metric. In this article, we describe share single-cell resolution 3D image dataset human carcinoma spheroids imaged using light-sheet fluorescence microscope. The contains 90 derived from 3 (i.e. T-47D, 5-8F, Huh-7D12) cleared with 5 protocols, precisely ClearT, ClearT2, CUBIC, ScaleA2, Sucrose. To evaluate quality light penetration depth samples, all under same experimental conditions, labelling nuclei DRAQ5 stain Leica SP8 Digital LightSheet was annotated by 10 independent experts thus allows microscopy users qualitatively compare effects protocols on lines.
